La presente investigación tuvo como objetivo contribuir a la validación de la actividad antimalárica de una preparación tradicional (RT) a base de Curarea Toxicofera (Wedd). Barneby and Krukoff, utilizada como antimalárico por la comunidad indígena Ciudad Hitoma del Amazonas Colombiano, adicionalmente se planteó evaluar el tipo de interacción que se presenta tras la administración conjunta con cloroquina (CQ) y artesunato (AS) dos medicamentos de uso común en la enfermedad. Con el fin de valorar la actividad antiplasmodial de RT, CQ y AS, se empleó en el modelo in vitro, el ensayo de inhibición de la invasión y desarrollo del parásito, empleando la cepa FCR-3 de Plasmodium falciparum y para evaluar la actividad antimalárica se utilizó el modelo de malaria murina “Test de Peters” cepa P. berghei ANKA los anteriores modelos farmacológicos se encuentran actualmente optimizados por el grupo de investigación FaMeTra. Para la evaluación de las interacciones entre el RT y CQ y entre el RT y AS, se emplearon los mismos modelos farmacológicos con algunas adaptaciones requeridas para las pruebas. Los resultados de las combinaciones dieron como respuesta in vitro, sinergismo para RT-CQ y aditividad para RT-AS; los resultados in vivo dieron como interacción aditividad para las dos combinaciones. Siendo un resultado positivo y promisorio tanto para el RT como posible antimalárico, como para la combinación de este con los fármacos mencionados.Summary
Abstract. The research had as an objective to contribute to the validation of the antimalarial activity of a traditional preparation based of Curarea Toxicofera (Wedd). Barneby and Krukoff (RT), used as antimalarial by the indigenous community ciudad Hitoma from the Colombian amazon. In addition, it was raised the type of interaction that was developed when it was delivered in companion of chloroquine (CQ) and artesunate (AS), two medicines commonly used in the treatment of malaria. In order to evaluate the antiplasmodial of RT, CQ and AS the in vitro model, inhibition of the invasion essay and the development of the parasite using the strain FCR-3 of Plasmodium falciparum and to evaluate the antimalarial activity the model Murina “Peters test”, strain P.Berghel ANKA was used. The previous pharmacological models were optimized by the research group FaMeTra. For the evaluation of the interaction between RT and CQ, and between the RT and AS, the previous pharmacological models were used, with some required adaptations for the tests. The results of the combinations gave as an in vitro result, synergism for RT-CQ and additivity for RT-AS, the in vivo results gave as interaction additivity for both combinations. Being a positive and promissory result for both RT as a possible antimalarial, and for its combination with the mentioned drugsKeywords
