Estudio químico y evaluación de la actividad antihipertensiva y antiagregante plaquetaria del extracto de peridermis de Solanum tuberosum (SOLANACEAE)

El estudio fitoquímico llevado a cabo en la peridermis del tubérculo de la especie Solanum tuberosum variedad sabanera (Solanaceae), permitió aislar un compuesto con actividad antiagregante plaquetaria y otro con potencial efecto antihipertensivo, ambos obtenidos a partir del extracto etanólico empleando diferentes técnicas cromatográficas. A partir del extracto etanólico, se obtuvieron 10 fracciones de polaridad creciente que fueron sometidas a ensayos de agregación plaquetaria por medio de la técnica turbidimétrica de Born, empleando plaquetas humanas inducidas con los agonistas (Colágeno, ácido Araquidónico, Epinefrina y ADP). Como resultado, se obtuvo a través de un análisis estadístico que las fracciones EPS-2 y EPS-7 presentaban diferencias frente al control (DMSO), indicando la posible presencia de metabolitos con actividad antiagregante plaquetaria. Posteriormente, las fracciones EPS-2 y EPS-7 fueron fraccionadas utilizando diferentes técnicas cromatográficas para purificar los compuestos SP-1 y SP-2 respectivamente. Estos compuestos fueron sometidos a los ensayos de agregación plaquetaria, obteniendo que el compuesto SP-2 identificado por RMN como Cafeato de etilo, presenta actividad antiagregante plaquetaria frente a los cuatro agonistas empleados, y se comporta de manera similar al patrón ácido acetilsalicílico (ASA). En una segunda fase, se construyó una curva de concentración del compuesto SP-2 – porcentaje de inhibición de la agregación plaquetaria, para obtener los valores de CI50 (µg/mL) frente a cada agonista: Epinefrina (59,47), APD (61,05), Colágeno (63,23) y Ácido araquidónico (62,75). Adicionalmente, y considerando reportes etnobotánicos e investigaciones recientes llevadas a cabo en la Universidad Nacional de Colombia, se evaluó el efecto hipotensor en rata desmedulada de las fracciones apolares EPS-1, EPS-2 y EPS-3 obtenidas del extracto etanólico. Los resultados de esta investigación, permiten evidenciar que la fracción de tipo apolar EPS-2, tiene efecto antihipertensivo en rata desmedulada estimulada con noradrenalina. Sin embargo, también se pudo verificar que esta actividad no es atribuida a los ésteres de ácido graso presentes en este tipo de fracciones como sugieren estudios anteriores. En consecuencia, el efecto antihipertensivo de esta fracción, puede ser ocasionado por otro tipo de compuestos como el Ferulato de tetradecilo, aislado e identificado por RMN de la fracción EPS-2. En conclusión, esta investigación resalta el potencial farmacológico del extracto de peridermis de S. tuberosum variedad sabanera, como fuente de metabolitos activos para el tratamiento de afecciones cardiovasculares.Summary
Abstract: The phytochemical study carried out on the tuber periderm of the species Solanum tuberosum variety sabanera (Solanaceae) allowed the chromatographic isolation of a compound with platelet antiaggregant activity and another one with potential antihypertensive effect, both of them obtained from the ethanolic extract. From the ethanolic extract, 10 fractions of increasing polarity were obtained. Their biological evaluation was performed with Born’s turbidimetric method and different agonists-induced platelet aggregation (Collagen, Arachidonic acid, Epinephrine and ADP). After statistical analysis, it EPS-2 and EPS-7 fractions showed significant differences in comparison with the control (DMSO), which indicates the possible presence of metabolites with platelet antiaggregant activity. Subsequently, the fractionation of EPS-2 and EPS-7 leaded to the purification of the compounds SP-1 and SP-2, respectively. These compouns were also subjected to the platelet aggregation test. The SP-2, identified by NMR as Ethyl Cafeate inhibited platelet aggregation induced by the four used agonists, and behaved similarly to the standard acetylsalicylic acid (ASA). In a second phase of the project, a dose-response curve for SP2 was generated. The stimated IC50 values (μg/mL) against each agonist-induced platelet aggregation were Epinephrine (59,47), APD (61,05), Collagen (63,23) and arachidonic acid (62,75). On the other hand, considering ethnobotanical reports and recent research carried out at the National University of Colombia, the hypotensive effect of the apolar fractions EPS-1, EPS-2 and EPS-3 obtained from the ethanolic extract was evaluated in pithed rat. The results showed that the apolar EPS-2 fraction has antihypertensive effect in a pithed rat stimulated with noradrenaline. In addition, this activity was not attributed to the fatty acid esters present in this type of fractions, as previous studies suggest. Conversely, the antihypertensive effect of this fraction might be caused by other types of compounds such as tetradecyl ferulate, which was isolated from the EPS-2 fraction.and identified through NMR. In conclusion, this research project highlights the pharmacological potential of the peridermium extract of S. tuberosum variety sabanera as a source of active metabolites for the treatment of cardiovascular diseases.Keywords
