La glándula tiroides regula la tasa metabólica de muchos órganos. El yodo es un componente esencial de las hormonas tiroideas, la T3 (triyodotironina) y la T4 (tetrayodotironina o tiroxina). La amiodarona es un agente poderoso y ampliamente usado como antiarrítmico pero efectos secundarios en esta glándula han sido reportados:, hipotiroidismo como hipertiroidismo con intervalos generales estimados entre 2% y el 24%, siendo el intervalo más común entre el 14% al 18%. Dos hipótesis pueden explicar estas disfunciones en la glándula tiroides: la primera, se puede relacionar con el aumento en la concentración de yodo debido al alto contenido de este halógeno presente en la molécula (37% de su peso molecular) (efecto Wolff-Chaikoff); la segunda, en donde el efecto de la amiodarona es directo e independiente de la concentración de yodo. En este trabajo nosotros estudiamos si hay modificaciones en el transporte del yoduro del tirocito por exposición directa a la amiodarona en folículos tiroideos de ratón in vitro y el papel de sus concentraciones altas de yoduro. Se analizó el estado redox, el transporte de yoduro y la localización de NIS, co-transportador responsable de la entrada de yoduro al tirocito dando una mejor comprensión de las alteraciones que se han reportado. / Abstract. The thyroid gland regulates the rate of metabolism of many organs. The iodine is an essential component of thyroid hormones, T3 (tri-iodothyronine) and T4 (tetra-iodothyronine or thyroxine). Amiodarone is a powerful and widely-used anthyarritmic agent, but side effects are reported: hypothyroidism as hyperthyroidism with a range estimated between 2% and 24%, most commonly in the range of 14–18%. Two hypotheses could explain these dysfunctions of the thyroid: firstly, it could relate to chronic high iodine concentrations due to the high content of this halogen present in the molecule (37% of it molecular weight) (Wolff-Chaikoff effect); secondly, the amiodarone effect is direct and independent of the iodine concentration. In this work, we studied if there are modifications in the iodine transport in the thyrocytes by direct exposition to amiodarone in thyroid follicles of mice in vitro and the role of its high iodide concentrations. The redox-state, the iodide transport and the localization of NIS, symporter responsible of the iodide entrance to the thyrocyte, were analyzed, let a better understanding of the alterations have been reported.Keywords
