Identificación de polimorfismos en el gen gyrA de Pseudomonas aeruginosa de pacientes con fibrosis quística y otras patologías en la ciudad de Cartagena (Colombia)

La fibrosis quística (FQ) es una enfermedad genética debida a mutaciones en el gen CFTR que codifica un canal de cloruro. Los pacientes con FQ expresan un canal no funcional en la membrana de las células epiteliales, y son susceptibles a infecciones pulmonares crónicas por P. aeruginosa. En este trabajo, se analizó la resistencia a ciprofloxacina de 60 aislados clínicos de P. aeruginosa: 30 de pacientes FQ y 30 de pacientes con otras patologías (no-FQ), y se secuenció la región QRDR del gen gyrA para identificar polimorfismos relacionados con resistencia a quinolonas. Se encontraron más aislados resistentes a ciprofloxacina en el grupo FQ (19/30) comparado con el grupo no-FQ (4/30) (p0.001). Se identificaron polimorfismos en ambos grupos: Thr-83Ile, Asp87Asn, y Asp87Gly, entre otras. El número de aislados con resistencia a ciprofloxacina con mutaciones en gyrA fue significativamente mayor en el grupo FQ (11/24) comparado con el grupo no-FQ (3/21) (p0,05), sugiriendo que la resistencia a ciprofloxacina en este grupo podría deberse a las mutaciones encontradas. / Abstract. Cystic fibrosis (CF) is a genetic disease due to mutations in CFTR gene encoding for a chloride channel. CF patients express a non-functional channel in epithelial cell membranes and they are susceptible to chronic lung infections by P. aeruginosa. In this work, resistance to ciprofloxacin of 60 P. aeruginosa clinical isolates was analyzed: 30 isolates from CF patients and 30 from other pathologies (non-CF), and the QRDR region of GyrA gene was sequenced to identify polymorphisms related to quinolone resistance. More ciprofloxacin-resistant isolates were found in the CF group (19/30) compared to non-CF group (4/30) (p0.001). Polymorphisms in both groups of isolates were identified: Thr-83Ile, Asp87Asn, and Asp87Gly, among others. The number of ciprofloxacin-resistant isolates with mutations in gyrA was significantly higher in the CF-group (11/24) compared to non-CF group (3/21) (p0.05), suggesting that the ciprofloxacin-resistance in this group might be due to the mutations identified.Keywords
