La caña de azúcar (Saccharum spp.) pertenece a la familia Poaceae y representa uno de los cultivos de mayor importancia económica debido a su alta acumulación de sacarosa, contabilizando cerca del 70% de la producción mundial, alta eficiencia en producción de biomasa, producción de biocombustibles, de primera y segunda generación, y cogeneración de energía. Se cultiva en áreas tropicales y subtropicales del mundo. En Colombia el cultivo de caña para la producción de azúcar ocupa un área de 218.311 ha produciendo en promedio 114.6 toneladas de caña por hectárea. El proceso de transformación genética en plantas permite introducir genes asociados a características que mejoren la calidad de un cultivo. Los promotores hacen parte de un gen y tiene la función de acoplar la maquinaria transcripcional para asegurar su expresión. Actualmente el proceso de transformación genética en caña de azúcar no dispone de un promotor obtenido a partir del genoma de la caña, estable a través del tiempo. Por lo tanto el objetivo de éste estudio fue aislar un promotor propio del genoma de la caña y evaluarlo en la inducción del gen reportero uidA (Gus). A partir del ADN de la variedad CC 01-678 se amplificaron dos fragmentos con tamaños de 1282 pares de bases (pb) (AY699) y 1303 pb (AY700), correspondientes al promotor del gen sacarosa sintetasa (SuSy). Por otro lado el promotor Ubiquitina de maíz, con un tamaño de 2000 pb, se usó como control y se obtuvo por medio de PCR a partir del plásmido pUbi/1SD. Tres construcciones genéticas denominadas pCAY699, pC-AY700 y pC-Ubi se obtuvieron reemplazando el promotor CaMV35S presente en el vector pCambia 1305.2 por los dos fragmentos aislados del gen SuSy así como por el promotor Ubiquitina. El análisis histoquímico de expresión transitoria del gen reportero uidA, dirigida por los diferentes promotores, se realizó utilizando callo embriogénico de la variedad CC 84-75. La mayor expresión se observó con el promotor Ubiquitina, contabilizando en promedio 406 ± 9.77 puntos azules, mientras que con el promotor CaMV35S y AY700 sólo se observaron 46 ± 3.48 y 4 ± 1.33 respectivamenteSummary
//Abstract: / Sugarcane (Saccharum spp.) belongs to the family Poaceae and is one of the most economically important crops because of its high accumulation of sucrose, accounting for about 70% of global output, high efficiency in biomass production, production biofuels, of first and second generation, and energy cogeneration. It is cultivated in tropical and subtropical countries around the world. In Colombia, the cultivation of sugarcane for sugar production occupies an area of 218,311 ha, produced an average of 114.6 tonnes of cane per hectare. The process of genetic transformation in plants allows the introduction of genes associated with characteristics that improve the quality of a crop. The promoters are part of a gene and have the function of coupling the transcriptional machinery to ensure its expression. Currently the genetic transformation of sugarcane does not have a promoter obtained from the sugarcane genome, stable over time. Therefore, the objective of this study was to isolate a promoter from the sugarcane genome and evaluate the induction of the reporter gene uidA (Gus). From the DNA of the sugarcane variety CC 01-678, two fragments with sizes of 1282 base pairs (bp) (AY699) and 1303 bp (AY700) were amplified from the sucrose synthase gene promoter (SuSy). Furthermore, maize Ubiquitin promoter with a size of 2000 bp, was used as control and was obtained by PCR from plasmid pUbi/1SD. Three genetic constructs called pC-AY699, pC-AY700 and pC-Ubi, were obtained by replacing the CaMV35S promoter present in the vector pCambia 1305.2 by each of the two fragments isolated from the SuSy gene as well as the ubiquitin promoter. Histochemical analysis of transient expression of uidA reporter gene, directed by the different promoters, was performed using embryogenic callus of variety CC 84-75. The highest expression was observed using the ubiquitin promoter, accounting on average 406 ± 9.77 blue dots, while using the CaMV35S and AY700 only 46 ± 3.48 and 4 ± 1.33 blue dots was observed respectively.Keywords
