En los últimos años, la Organización Mundial de la Salud se ha preocupado por la rápida aparición de microorganismos resistentes a los antimicrobianos; estos son el resultado tanto de la presión selectiva por parte de los fármacos usados, como del uso irracional de los mismos, por esta razón es importante buscar nuevas estrategias para combatir las cepas resistentes. Las investigaciones en el campo de los antimicrobianos están dirigidas al desarrollo y búsqueda de nuevas moléculas. En este campo se destacan los péptidos, los cuales se han perfilado como una familia de moléculas antimicrobianas denominadas (PAMs). El estudio de los péptidos antimicrobianos tiene como finalidad mejorar la actividad y entender su mecanismo de acción. El objetivo de este trabajo fue sintetizar y determinar la actividad antimicrobiana de pseudopéptidos (análogos peptídicos amida reducida) del fragmento C-terminal de la protamina; estas moléculas mostraron características heterogéneas respecto a la especificidad frente a cepas Gram positivas, Gram negativas y tiempos de vida media, pero en general las modificaciones disminuyeron la actividad hemolítica sin afectar la estructura nativa. Esta estrategia permite modular la actividad antimicrobiana del péptido nativo; sin embargo, es necesario complementar este tipo de estudio con pruebas específicas y determinar si dos o más modificaciones simultáneas en una molécula podrían actuar de forma sinérgica para mejorar la actividad contra un microorganismo determinado.Resumen
Abstract. In recent years, the World Health Organization has been concerned about the rapid emergence of resistant organisms to antimicrobials, the resistant organisms are the result of both selective pressure of the drugs used and the irrational use thereof, and for this reason it is important to seek new strategies to combat resistant strains. Research in the field of antimicrobials is directed to the development and research of new molecules. In this field the peptides have emerged as a family of antimicrobial molecules called (PAMs). The study of antimicrobial peptides intended to improve the activity and understand its mechanism of action. The aim of this work was to synthesize and establish the antimicrobial activity of pseudopeptides (reduced amide peptide analogs) of the C-terminal fragment of protamine, these molecules showed heterogeneous characteristics for specificity against strains Gram positive, Gram negative and times half life, but in general the modifications reduced the hemolytic activity without affecting the native structure.
