Síntesis de N- glucosilaminoácidos empleando el anhídrido cíclico del ácido propilfosfónico T3P® como agente acoplante

dc.description.abstract | Se realizó la síntesis y caracterización por Resonancia Magnética Nuclear y cromatografía RP-HPLC de los N-glucosilaminoácidos: Fmoc-Phe-O-(2,3,4,6-tetra-O-Acetil-β-D-N-glucopiranosil), Fmoc-Leu-O-(2,3,4,6-tetra-O-Acetil-β-D-N-glucopiranosil), Fmoc-Lys-(Boc)-O-(2,3,4,6-tetra-O-Acetil-β-D-N-glucopiranosil), Fmoc-Asp-(OtBu)-O-(2,3,4,6-tetra-O-Acetil-β-D-N-glucopiranosil), L-Phe-O-β-1N(D-glucopiranosil), L-Leu-O-β-1N(D-glucopiranosil), L-Lys-O-β-1N(D-glucopiranosil) y Asp-O-β-1N(D-glucopiranosil). La síntesis de estos N-glucosilaminoácidos no ha sido reportada hasta el momento. Se diseñó e implementó la ruta sintética utilizando metodologías más amigables con el medio ambiente. Fue posible identificar los puntos criticos de la ruta sintética y buscar estrategias de mejora. Los resultados obtenidos muestran que la metodología empleada puede ser considerada como una alternativa viable y novedosa para la obtención de glucoconjugados aminoácido-carbohidrato. Este trabajo contribuirá al diseño y obtención de N-glucosilaminoácidos que pueden ser considerados para estudios de la inhibición de la adhesión de Pasteurella Multocida a tejido epitelial de conejo. |
dc.description.abstract | Abstract: The synthesis and characterization by Nuclear Magnetic Resonance and RP-HPLC chromatography of N-glycosyl amino acids were perfomed. The N-glycosyl amino acids obtained were: Fmoc-Phe-(2,3,4,6-tetra-O-acetyl-β-D-N-glucopyranose), Fmoc-Leu--(2,3,4,6-tetra-O-acetyl-β-D-N-glucopyranose), Fmoc-Lys-(Boc)-O-(2,3,4,6-tetra-O-acetyl-β-D-N-glucopyranose), Fmoc-Asp-(OtBu)-O-(2,3,4,6-tetra-O-Acetil-β-D-Nglucopyranose), L-Phe-O-β-1N(D-glucopyranose), L-Leu-O-β-1N(D-glucopyranose), LLys-O-β-1N(D-glucopyranose) and Asp-O-β-1N(Dglucopyranose. Until now, the synthesis of these N-glucosyl amino acids has not been reported in literature. The synthetic route was designed and performed using environmentally-friendlier methodologies. It was possible to identify the critical points in the synthetic route and to look for improvement strategies. The obtained results showed that the methodology carried out can be considered as a viable and novel alternative for the obtention of glicoconjugates amino acid-carbohydrate. The present work will contribute to the design and obtention of N-glucosyl amino acids that could be used to evaluate adhesion inhibition of Pasteurella multocida to rabbit epithelial tissue. |
