dc.description.abstract | La sericultura o producción de seda a partir de la cría del gusano de seda Bombyx mori L. (Bombycidae) es una actividad agroindustrial estratégica para el desarrollo socio-económico del departamento del Cauca, Colombia. Analizar la diversidad genética de B. mori es fundamental para la conservación y uso de sus recursos genéticos. El estudio evaluó 13 materiales japoneses, chinos e híbridos, del banco de germoplasma de B. mori de la Universidad del Cauca, ubicado en el departamento del Cauca, utilizando seis microsatélites (SSR) y la región COI. Fueron detectados 33 alelos en los loci SSR, con un rango entre 3 y 9, y un PIC entre 0,317 y 0,701, indicando que FL0933 y FI0619 son marcadores altamente informativos. En promedio, los valores de cada material para el número de alelos por locus (Npa), la heterocigosidad promedio entre loci observada (Hpo) y esperada (Hpe) fueron de 2,179, 0,347 y 0,321 respectivamente, indicando altos niveles de homocigosidad. Los híbridos presentaron valores superiores de Hpe y Npa, seguidos por los japoneses y chinos, señalando de mayor a menor el nivel de variabilidad genética presente en estos grupos poblacionales. El AMOVA y los índices de fijación no revelaron diferencias intrapoblacionales significativas (3%), señalando que los materiales están en equilibrio de Hardy-Weinberg, sin embargo, indicaron diferencias significativas entre los híbridos, japoneses y chinos (10%), así como entre los materiales que conforman estos grupos poblacionales (27%). El análisis de agrupamiento generó tres grupos, el I y el III confirman la separación por el origen geográfico, este último esta distanciado genéticamente del resto por valores promedio superiores al 0,4, debido a la presencia de seis alelos exclusivos en los materiales japoneses. La región COI presentó un sitio segregante y dos haplotipos que confirmaron la procedencia europea de los materiales, pero la variabilidad fue insuficiente para discriminarlos individualmente. Los hallazgos sugieren la disponibilidad de diversidad genética amplia y estructurada, distribuida principalmente en los grupos poblacionales híbrido y japonés, revelando además que estos últimos representan un grupo poblacional altamente diferenciado. |
dc.description.abstract | //Abstract: Sericulture, the production of silk from the breeding of the silkworm Bombyx mori L. (Bombycidae) is a strategic agro-industrial activity for the socio-economic development of the department of Cauca, Colombia. Analyzing the genetic diversity of B. mori is fundamental for the conservation and use of its genetic resources. The study evaluated 13 Japanese, Chinese and hybrid materials, from the B. mori germplasm bank of “Universidad del Cauca” located in the department of Cauca, using six microsatellites (SSR) and the COI region. Thirty-three alleles were detected in the SSR loci, with a range between 3 and 9, and a PIC between 0,317 and 0,701, indicating that FL0933 and FI0619 are highly informative markers. On average, the values of each material for the number of alleles per locus (Npa), the average heterozygosity observed (Hpo) and expected (Hpe) between loci were 2,179, 0,347 and 0,321 respectively, indicating high homozygosity levels. The hybrids presented superior values of Hpe and Npa, followed by the Japanese and Chinese materials, indicating from greater to lesser the level of genetic variability present in these population groups. The AMOVA and fixation indices did not reveal significant intrapopulation differences (3%), indicating that the materials are in Hardy-Weinberg equilibrium, however indicated significant differences between hybrids, Japanese and Chinese (10%), as well as between the materials that comprise these population groups (27%). The cluster analysis generated three groups, I and III confirm the separation by geographical origin, the latter is genetically distanced from the rest by average values higher than 0,4, due to the presence of six exclusive alleles in Japanese materials. The COI region presented a segregating site and two haplotypes that confirmed the European origin of the materials, but the variability was insufficient to discriminate them individually. The findings suggest the availability of broad and structured genetic diversity, distributed mainly in the hybrid and Japanese population groups, also revealing that the latter represent a highly differentiated population group. |
