dc.description.abstractThe genus Vibrio spp has gained relevance worldwide due to the increase in aquatic environments and causing clinical cases called vibriosis. Currently the distribution and abundance of this genus in Colombia is unknown. The objective of this work is to identify and genotypically characterize isolates of V. cholerae, V. parahaemolyticus, V. alginolyticus, V. fluvialis and V. vulnificus, recovered from environmental samples (bodies of water) and clinical samples (suspected cases for Vibrio spp.). To do this, the search for the 5 included species was carried out using multiplex PCR. In addition, the virulence genes (ctxA, tcp, nanH, hlyA), (tdh, trh), vfh and (vcgC or vcgE), in each species respectively, were screened. the clonal relationship between the species will be established using PFGE. 240 isolates were evaluated: 195 of environmental origin collected by INVEMAR in 2018 and 2019 and sent to the INS from 2010 to 2019, and 45 of clinical origin sent to the INS in the same period. Of the total, 168 were identified and classified as one of the 5 white species. At the prevailing environmental level, V. parahaemolyticus (n = 41) and V. alginolyticus (n = 28), while at the prevailing clinical level, V. cholerae No O1 / O139 (n = 29) and V. fluvialis (n = 23). Some isolates carried virulence genes such as: nanH (43%), hlyA (54.5%), tdh (16.4%), vfh (96%), vcgC (85.7%) or vcgE (14.2%). All statistical species have great genetic diversity with the presence of some circulating clones in the country. The presence and distribution of 5 potentially pathogenic Vibrio species in the country in environmental and clinical samples was confirmed, which could affect as a public health problem.
dc.description.abstractEl género Vibrio spp, ha cobrado relevancia a nivel mundial debido al incremento en ambientes acuáticos y causando casos clínicos denominados vibriosis. En la actualidad se desconoce la distribución y abundancia de este género en Colombia. El objetivo de este trabajo es identificar y caracterizar genotípicamente aislamientos de V. cholerae, V. parahaemolyticus, V. alginolyticus, V. fluvialis y V. vulnificus, recuperados de muestras ambientales (cuerpos de agua) y muestras clínicas (casos sospechosos para Vibrio spp.). Para ello se realiza búsqueda de las 5 especies mencionadas mediante PCR multiplex, adicionalmente se tamizaron los genes de virulencia (ctxA, tcp, nanH, hlyA), (tdh, trh), vfh y (vcgC o vcgE), en cada especie respectivamente y se estableció la relación clonal entre las especies mediante PFGE. Se evaluaron 240 aislamientos: 195 de origen ambiental recolectados por INVEMAR en 2018 y 2019 y remitidos al INS de 2010 a 2019, y 45 de origen clínico remitidos al INS en el mismo periodo. Del total, 168 se identificaron y clasificaron como alguna de las 5 especies blanco. A nivel ambiental predomino V. parahaemolyticus (n=41) y V. alginolyticus (n=28), mientras que a nivel clínico predomino V. cholerae No O1/O139 (n=29) y V. fluvialis (n=23). Algunos aislamientos fueron portadores de genes de virulencia como: nanH (43%), hlyA (54.5%), tdh (16,4%), vfh (96%), vcgC (85.7%) o vcgE (14.2%). Todas las especies mostraron gran diversidad genética con presencia de algunos clones circulantes en el país. Se confirmó la presencia y distribución de 5 especies potencialmente patogénicas de Vibrio en el país en muestras ambientales y clínicas, lo que podría considerarse como un problema de salud pública.
