dc.description.abstractUltraviolet radiation is one of the main causes of skin cancer associated with the generation of oxidative stress. Therefore, substances with antioxidant and / or photoprotective properties are used for its prevention, since on the one hand, they reduce oxidative stress and on the other hand they avoid the incidence of said radiation. Despite the use of traditional photoprotectors, skin cancer continues to grow, which makes it necessary to search for novel molecular entities with dual properties (antioxidant and photoprotective) that are potentially more effective. Everniastrum cirrhatum (Fr.) Hale (Parmeliaceae) a lichenized fungus from Sumapaz páramo, was subjected to study to isolate its chemical constituents and determine its biological activity in order to find said agents. From its acetonic extract, were isolated and identified: salazinic acid (1), pseudocifelarin A (2), atraric acid (3), atranorin (4) and usnic acid (5). Compounds 2 and 5 are reported here for the first time for the species. Their antioxidant activity was determined in-vitro by evaluating their free radical scavenging power (DPPH•), their iron-reducing power and their power to inhibit lipid peroxidation, while their photoprotective capacity was evaluated in-vitro against UVB and UVA radiation. Additionally, the lipophilicity and preliminary permeability of these compounds were calculated, considering they can be used as ingredients of formulations intended for the skin, which in turn is an organ that has a lipophilic nature. Compounds 1, 4 and 5 were dual agents with antioxidant properties that involved iron reduction and inhibition of lipid peroxidation, which also showed high photoprotective capacity against UVB and UVA radiations. Additionally, 2 was an agent with "very high" photoprotective capacity against UVA radiation. These compounds have adequate permeability to be delivered to the stratum corneum of the skin, therefore they could become active principles for the development of formulations aimed at the prevention of conditions associated with solar radiation, including skin cancer.
dc.description.abstractLa radiación ultravioleta constituye uno de los principales factores causantes de cáncer de piel asociado a la generación de estrés oxidativo en dicho órgano. Por ello, sustancias con propiedades antioxidantes y/o fotoprotectoras, se usan para su prevención ya que, por un lado, disminuyen el estrés oxidativo y por otro lado evitan la incidencia de dicha radiación. Pese al uso de fotoprotectores tradicionales, el cáncer de piel sigue en continuo crecimiento, lo cual hace necesaria la búsqueda de entidades moleculares novedosas con propiedades duales (antioxidantes y fotoprotectoras) que resulten potencialmente más eficaces. Everniastrum cirrhatum (Fr.) Hale (Parmeliaceae) un hongo liquenizado del páramo de Sumapaz, fue sometido a estudio para aislar sus constituyentes químicos y determinar su actividad biológica con la finalidad de encontrar dichos agentes. A partir de su extracto acetónico, se aislaron e identificaron a los compuestos: ácido salazínico (1), pseudocifelarina A (2), ácido atrárico (3), atranorina (4) y el ácido úsnico (5), de los cuales 2 y 5 son reportados por primera vez para la especie. La actividad antioxidante se determinó in-vitro evaluando su poder captador de radicales libres (DPPH•), su poder reductor férrico y su poder para inhibir la peroxidación lipídica, mientras que su capacidad fotoprotectora se evaluó in-vitro frente a la radiación UVB y UVA. Adicionalmente, se calculó la lipofilia y permeabilidad preliminar de los compuestos, considerando que estos pueden ser empleados como ingredientes de formulaciones destinadas a la piel, órgano que posee una naturaleza lipofílica. Los compuestos 1, 4 y 5 resultaron ser agentes duales con propiedades antioxidantes que involucran la reducción férrica y la inhibición de la peroxidación lipídica, que además poseen capacidad fotoprotectora alta en contra de la radiación UVB y UVA. Adicionalmente, 2 resultó ser un agente fotoprotector con “muy alta” capacidad fotoprotectora frente a la radiación UVA. Dichos compuestos poseen características de permeabilidad adecuada para se entregadas en el estrato córneo de la piel, por lo cual se podrían constituir en principios activos para el desarrollo de formulaciones destinadas a la prevención de padecimientos asociados a la radiación solar, entre ellos el cáncer de piel.
