Los estudios arqueogenómicos indican que el poblamiento de América sucedió hace aproximadamente 16.000 años antes del presente. Adicionalmente, Colombia fue un punto importante de entrada al continente suramericano, y cuenta con limitados estudios que describen su historia genético-poblacional. Las comunidades precolombinas del valle del río Magdalena son de interés a nivel de ADN antiguo, ya que, por su posición geográfica contribuyeron al desarrollo de procesos bioculturales y adaptativos que llevaron a migraciones, relaciones interpoblacionales, diversificación biológica y la colonización hasta el sur del continente. El objetivo de este estudio es analizar la estructura genética por vía matrilineal de restos óseos del valle del río Magdalena a través del análisis de la región control y genomas completos de ADN mitocondrial. Se obtuvieron datos de la región control en 11 muestras y 15 genomas mitocondriales de restos óseos precolombinos de la cuenca del río Magdalena, realizando posteriormente, asignación de haplogrupos mitocondriales, diversidad genética, filogenia matrilineal, y su relación entre los haplotipos encontrados a nivel inter e intrapoblacional, a través de los softwares Arlequín, Network y mtPhyl. Los linajes del ADNmt observados en este grupo poblacional pertenecen a los haplogrupos nativoamericanos A, B, C y D, se lograron definir 9 subhaplogrupos que están relacionados con otras poblaciones antiguas y contemporáneas de América o que, en otros casos, se encuentran aislados y restringidos a poblaciones colombianas y sus zonas fronterizas, las cuales revelan relaciones intra e interpoblacionales y sucesos de contacto importantes. En conclusión, este reporte de genomas mitocondriales precolombinos del Magdalena complementa las hipótesis arqueológicas del poblamiento suramericano, de los movimientos migracionales, de la diversidad genética y de la historia cultural de estas poblaciones evidenciando linajes fundadores y que se encuentran estrechamente ligados por línea materna con una alta diversidad genética. Archaeogenomic studies indicate that the peopling of America occurred approximately 16,000 years ago before the present. Furthermore, Colombia was an important entry point to the South American continent, and has limited studies that describe its genetic-population history. The pre-Columbian communities of the Magdalena River Valley are of interest at the level of ancient DNA, since, due to their geographical position, they contributed to the development of biocultural and adaptive processes that led to migrations, interpopulation relationships, biological diversification and colonization to the south of the continent. The aim of this study is to analyze the genetic structure through the matrilineal phylogeny of human skeletons from the Magdalena River Valley through the analysis of the control region and complete mitochondrial DNA genomes. Data from the control region were obtained in 11 samples and 15 mitochondrial genomes from pre-Columbian dental samples from the Magdalena River Valley, subsequently assigning mitochondrial haplogroups, genetic diversity, matrilineal phylogeny, and their relationship between the haplotypes found at the inter- and intrapopulation, through the Arlequín, Network and mtPhyl software. The mtDNA lineages observed in this population belong to the Native American haplogroups A, B, C and D, it was possible to define 9 subhaplogroups that are related to other ancient and contemporary populations of America and others, are isolated and restricted to Colombian populations and their border areas, which reveal intra- and inter-population relationships and important contact events. In conclusion, this report of pre-Columbian mitochondrial genomes from Magdalena complements the archaeological hypotheses of the South American settlement, migratory movements, genetic diversity and cultural history of these populations, evidencing founding lineages that are closely linked by maternal lineage with a high genetic diversity.Keywords
