Sustitución nucleofílica del grupo benzotriazolilo en el aminal 1,3-bis(benzotriazol-1-il-metil)-2,3 dihidrobenzimidazol

En esta investigación se mejoró el rendimiento de la síntesis del aminal 1,3- bis(benzotriazol-1-il-metil)-2,3-dihidrobenzimidazol haciendo modificaciones a la metodología de síntesis tricomponente propuesta por Katritzky y colaboradores. Se logró establecer que los productos de la reacción entre o-fenilendiamina, 1H benzotriazol y formaldehído corresponden a una mezcla de tres isómeros debido a la tautomería del benzotriazol en solución. Esta mezcla de isómeros fue caracterizada por RMN 1H. y mediante un proceso de recristalización en acetato de etilo, fue posible separar y caracterizar por RMN 1H, 13C y Rayos X el isómero mayoritario correspondiente a 1,3-bis(benzotriazol-1-il-metil)-2,3- dihidrobenzimidazol. Finalmente la reactividad del aminal 1,3-bis(benzotriazol-1-il-metil)-2,3- dihidrobenzimidazol fue evaluada frente a ión cianuro como nucleofilo, logrando la doble sustitución de los grupos benzotriazolilo en un medio de reacción polar prótico como metanol a temperatura ambiente, por otra parte se estableció que a temperatura de reflujo del metanol no solo conduce al mismo producto sino que además se generan nuevos productos que no necesariamente corresponden a productos de sustitución. / Abstract. In this research there was improved the synthesis performance of the aminal 1,3- bis (benzotriazole-1-il-metil)-2,3-dihidrobenzimidazol doing modifications to the methodology about the synthesis of three – components proposed by Katritzky et. al. It was possible to establish that the reaction`s products between o-phenylenediamine, 1H-benzotriazole and formaldehyde correspond to an isomeric mixture of three, because of the tautomerism of the benzotriazole in solution. This isomeric mixture, was characterized by RMN 1H; and by means of a recrystallization process in ethyl acetate it was possible to separate and characterize by RMN 1H, 13C and X Ray the major isomer, corresponding to 1,3-bis(benzotriazol-1-yl-methyl -) -2,3-dihydrobenzimidazole. Finally the reactivity of the aminal 1,3-bis(benzotriazol-1-yl-methyl)-2,3- dihydrobenzimidazole was evaluated opposite to cyanide ion as a nucleophile achieving the double substitution of the benzotriazolyl groups in a polar protic reaction medium as methanol in room temperature, on the other hand, it was established that in a reflux methanol temperature not only leads to the same product, but also new products are also generated that not necessarily correspond to substitution productsPalabras clave
