Spongospora subterranea f.sp. subterranea (Sss) causa la sarna polvosa de la papa, enfermedad que se ha expandido a todos los países productores de papa. En Colombia éste patógeno se reportó por primera vez en 1965 y desde entonces se registra en todas las zonas paperas afectando la apariencia y calidad del tubérculo por la presencia de pústulas y/o disminuyendo la producción por la formación de agallas en las raíces; a pesar de esto aún no existe un manejo efectivo de la enfermedad. El objetivo de ésta investigación fue analizar la interacción S. tuberosum Grupo Phureja (StP) y S. subterranea f.sp. subterranea en términos de especificidad por órgano de infección. Se estableció un ensayo con suelo naturalmente infestado bajo condiciones de campo en el primer semestre del año 2012, se evaluó resistencia y susceptibilidad en raíz y/o tubérculo en cuarenta y nueve genotipos de StP. Cuando el órgano evaluado mostraba la sintomatología típica de la enfermedad se tomaron muestras que se utilizaron para caracterizar al patógeno mediante PCR utilizando primers específicos para Sss que amplifican la región ITS 1-5.8 S-ITS 2 del ADNr. De los 49 genotipos, 32 presentaron agallas en raíz, 11 presentaron pústulas en tubérculo y 9 presentaron infección en ambos órganos. Se confirmó la especificidad del patógeno por órgano de infección. Se reporta que en las accesiones con síntomas en raíz y en tubérculo se identificaron las dos variantes del patógeno en una misma planta. Así también se encontraron mutaciones puntuales en la secuencia de nucleótidos corroborando que en Colombia la población de Sss es más variable genéticamente que las poblaciones del patógeno de otros países. Spongospora subterranea f.sp. subterranea (Sss) causes powdery scab of potato disease has spread to all potato producing countries. In Colombia Sss was reported in 1965 and since then is recorded in all areas affecting the appearance of tuber and quality by the presence of pustules and / or diminution the production by the formation of galls on roots, although this has not there is an effective management of the disease. The purpose of this study was to analyze the interaction S. tuberosum Grup (StP) and S. subterranea f.sp. subterranea in terms of organ specificity for infection. A trial was established with naturally infested soil under field conditions in the first semester of 2012, resistance and susceptibility was evaluated in root and / or tuber forty nine accessions of StP belonging to the National Collection of StP National University of Colombia. When the structure assessed showed typical symptoms of the disease were sampled that were used to characterize the pathogen by PCR using specific primers that amplify Sss - 5.8S – ITS 1 rDNA ITS 2. Of the 49 accessions, 33 had root galls, 11 had pustules on tuber and 9 had infection in both organs. Specificity confirmed by organ pathogen infection.
