dc.description.abstract | La soya es uno de los cultivos más importantes a nivel mundial. La productividad de este cultivo ha aumentado sustancialmente a partir de la adopción de líneas transgénicas con tolerancia a herbicidas, sin embargo, estas líneas se han desarrollado para regiones templadas. Por esta razón la implementación de soya tolerante a herbicidas en Colombia requiere la transformación de variedades nacionales. Uno de los aspectos más importantes en este proceso es el desarrollo de constructos génicos que expresen la característica de interés, los cuales tradicionalmente se han desarrollado mediante técnicas de biología molecular convencionales. Sin embargo, gracias a las herramientas de la bioinformática y la biología sintética, es posible realizar el diseño in silico del constructo y contratar la síntesis del mismo, permitiendo realizar modificaciones en la secuencia como optimizaciones de uso codónico, las cuales mejoran significativamente los niveles de expresión de la proteína. En este trabajo se diseñaron y evaluaron versiones semisintéticas de genes que confieren tolerancia a glifosato y glufosinato de amonio, como una fase inicial para la obtención de variedades nacionales de soya tolerantes a herbicidas. |
dc.description.abstract | Abstract. The Soybean crop is one of the most important in the world. The productivity of this crop has increased substantially since the adoption of tolerant herbicide transgenic lines. However these lines were developed for temperate regions, for this reason, the successful introduction of herbicide tolerance soybean in Colombia could require genetic transformation of the national varieties. One of the most important issues about this process is the development of genetic constructs that express the characteristic of interest, which have been developed by techniques of conventional molecular biology. However thanks to bioinformatic and synthetic biology tools, it is possible to make the in silico design of the construct and hiring its synthesis, allowing to perform sequence modifications like codon usage, which significatively improve the protein expression. In this work, there were designed and assessed semi-synthetic gene versions that confer glyphosate and glufosinate tolerance, as an initial phase for the obtention of national soybean varieties with herbicide tolerance. |
