dc.description.abstract | Canine Transmissible Venereal Tumor (CTVT) is a clonally transmissible neoplasm characterized by cytomorphological heterogeneity and variable responses to chemotherapy. This preliminary study used RNA-Seq data from 10 CTVT samples to explore transcriptomic differences among cytomorphological subtypes (plasmacytoid, lymphocytoid, and mixed) and among different chemotherapy responses (complete response, partial response, and resistance), through differential gene expression analysis, functional enrichment, and gene co-expression network analyses. Following a literature review, experimental findings are presented in two chapters: The first chapter explores gene expression patterns linked to cytomorphological subtypes, revealing that variations in antigen presentation and immune evasion pathways may underlie the distinct biological behaviors observed across these subtypes. The second chapter examines transcriptomic differences related to chemotherapy outcomes, identifying distinct molecular landscapes related with immune activation and mitochondrial apoptosis regulators. These findings provide initial insights into potential mechanisms driving tumor progression and chemoresistance in CTVT, suggesting candidate biomarkers and therapeutic targets. However, validation in larger cohorts and additional functional studies are required to substantiate these associations and elucidate their clinical significance. | eng |
dc.description.abstract | El tumor venéreo transmisible canino (CTVT, por sus siglas en inglés) es una neoplasia de transmisión clonal, caracterizada por marcada heterogeneidad citomorfológica y respuesta variable a la quimioterapia. En este estudio preliminar se utilizaron datos de RNA-Seq de 10 muestras de CTVT para explorar las diferencias transcriptómicas entre distintos subtipos citomorfológicos (plasmocitoide, linfocitoide y mixto) y entre diferentes respuestas a la quimioterapia con vincristina (respuesta completa, respuesta parcial y resistencia), mediante análisis de expresión génica diferencial, enriquecimiento funcional y redes de coexpresión génica. Tras una revisión de literatura exhaustiva, los hallazgos experimentales se presentan en dos capítulos. El primer capítulo analiza los patrones de expresión génica asociados a los subtipos citomorfológicos, evidenciando que variaciones en vías de presentación de antígenos y evasión inmune podrían sustentar los comportamientos biológicos distintivos observados entre ellos. El segundo capítulo examina las diferencias transcriptómicas relacionadas con respuesta a la quimioterapia, identificando perfiles moleculares diferenciados asociados con activación inmune y regulación de apoptosis mitocondrial. Estos resultados ofrecen una visión preliminar de posibles mecanismos que impulsan la progresión tumoral y la quimiorresistencia en CTVT, y sugieren potenciales biomarcadores y nuevos blancos terapéuticos. No obstante, se requiere la validación de estos hallazgos en cohortes más amplias y mediante estudios funcionales adicionales para confirmar las asociaciones observadas y esclarecer su relevancia clínica.
