dc.description.abstractEl hongo macromiceto Ganoderma lucidum, es una fuente de compuestos bioactivos con actividades biológicas científicamente comprobadas. A pesar de esto, hay pocos estudios enfocados a estudiar el potencial bioquímico de las cepas de este hongo comercializadas en nuestro país. Es por ello por lo que en esta investigación se presentan los resultados del estudio de la capacidad antioxidante y la actividad inhibitoria de AGEs in vitro, así como la presencia de lectinas con actividad aglutinante, de dos cepas de este hongo cultivadas en Colombia de manera tradicional a escala industrial (EUA y BEL). Para ello se realizaron extractos empleando solventes de diferente polaridad y se evaluaron por métodos espectrofotométricos, la capacidad antioxidante y la inhibición de la formación de productos avanzados de glicosilación AGEs. Este análisis se complementó con un estudio dirigido a las potenciales moléculas bioactivas, usando determinación de metabolitos por métodos espectrofotométricos, perfilado cromatográfico (HPLC-DAD y CG-MS) y análisis estadísticos multivariados (PLS-DA). Finalmente, se evaluó en un modelo in vitro la presencia de lectinas midiendo la aglutinación de eritrocitos a partir de extractos proteicos de las dos cepas en estudio. Se encontró que los extractos etanólicos y en diclorometano de las dos cepas, presentan actividad antioxidante e inhibitoria de AGE. Sin embargo, la cepa BEL presentó los mayores niveles de dichas actividades; el análisis cromatográfico y estadístico permitió encontrar qué, diferentes metabolitos del tipo triterpenos, están relacionados con el potencial antioxidante e inhibitorio de AGEs. Por otro lado, se encontró que en esta misma cepa se presenta importante actividad antiaglutinante; la purificación y caracterización de las lectinas presentes permitieron determinar que en la cepa BEL, se presentan lectinas con propiedades bioquímicas que le otorgan potencial para ser usadas en la industria de alimentos. El presente estudio contribuye al conocimiento de la composición bioquímica del hongo G. lucidum y confirma su actividad biológica in vitro en el micelio producido en Colombia. dc.description.abstractThe mushroom fungus Ganoderma lucidum is a source of bioactive compounds with scientifically proven biological activities. Despite this, there are few studies focused on studying the biochemical potential of the strains of this fungus commercialized in our country. For this reason, this research presents the results of the study of the antioxidant capacity and the inhibitory activity of AGEs in vitro, as well as the presence of lectins with binding activity, of two strains of this fungus traditionally cultivated in Colombia on an industrial scale (EUA and BEL). For this purpose, extracts were made, using solvents of different polarity, and were evaluated by spectrophotometric methods, the antioxidant capacity and the inhibition of the formation of advanced AGEs glycosylation products. This analysis was complemented with a study aimed at potential bioactive molecules, using metabolite determination by spectrophotometric methods, chromatographic profiling (HPLC-DAD and GC-MS) and multivariate statistical analysis (PLS-DA). Finally, the presence of lectins was evaluated in an in vitro model by measuring the agglutination of erythrocytes from protein extracts of the two strains under study. It was found that the ethanolic and dichloromethane extracts of the two strains showed antioxidant and AGE inhibitory activity. However, the BEL strain presented the highest levels of these activities; the chromatographic and statistical analysis allowed finding that different metabolites of the triterpene type are related to the antioxidant and AGE inhibitory potential. On the other hand, it was found that this strain has an important antiagglutinating activity; the purification and characterization of the lectins present allowed determining that the BEL strain has lectins with biochemical properties that give it the potential to be used in the industry. The present study contributes to the knowledge of the biochemical composition of the fungus G. lucidum and confirms it is in vitro biological activity in the mycelium produced in Colombia.eng
