Evaluación de algunos métodos tradicionales de identificación para detectar Salmonella spp. y Staphylococcus aureus coagulasa positiva en leche

dc.description.abstractLa leche es una matriz alimenticia con gran contenido nutricional, lo que estimula el crecimiento de bacterias como Salmonella Typhimurium y Staphylococcus aureus de alto impacto en la seguridad alimentaria, además, con una alta prevalencia en la leche, por lo que es necesario establecer metodologías para su detección. En este estudio se inocularon muestras de leche con estos microorganismos para establecer la capacidad del método horizontal, inmunológico (ELISA) y molecular (PCR) para detectarlos, para ello, se evaluó la detección del método horizontal inoculando simultáneamente S. Typhimurium.y S. aureus en leche cruda y ultrapasteurizada (UHT), también se realizaron pruebas inoculando leche UHT con estos dos microorganismos y la Bacteria ácido láctica (BAL) de la especie Lactococcus lactis para evaluar la detección de ellos en presencia de una bacteria que puede hacer parte de la microbiota de la leche. Con los métodos horizontal y ELISA se identificó S. Typhimurium en todas las muestras, mientras que la PCR no logró su identificación. Por otro lado, S. aureus solo se identificó por el método horizontal con un 90% de detección. En la inoculación simultánea se compararon los resultados con el recuento de UFC/mL de S. aureus, debido a que para S. Typhimurium no se requiere recuento sino ausencia/presencia. Se encontró que en la muestra de leche UHT inoculada con S. Typhimurium.y S. aureus se observó un mayor recuento que en la leche cruda, lo que indica que la microbiota presente en la leche cruda pudo ejercer un efecto inhibidor sobre el crecimiento de S. aureus; en contraste, la leche UHT inoculada adicionalmente con L. lactis, tuvo un mayor crecimiento que en todas las inoculaciones, lo que sugiere algún tipo de interacción entre los tres microorganismos. dc.description.abstractMilk is a nutritional matrix with high nutritional content, which promotes the growth of bacteria such as Salmonella Typhimurium and Staphylococcus aureus, having a significant impact on food safety. Moreover, these bacteria have a high prevalence in milk, making it necessary to establish methodologies for their detection. In this study, milk samples were inoculated with these microorganisms to assess the capability of the horizontal, immunological (ELISA), and molecular (PCR) methods for detection. The horizontal method detection was evaluated by simultaneously inoculating S. Typhimurium and S. aureus in raw and ultra-pasteurized milk (UHT). Tests were also conducted by inoculating UHT milk with these two microorganisms and lactic acid bacteria (LAB) of the Lactococcus lactis species to evaluate their detection in the presence of bacteria that may be part of milk's microbiota. Using the horizontal and ELISA methods, S. Typhimurium was identified in all samples, while PCR failed to identify it. On the other hand, S. aureus was only identified by the horizontal method with a detection rate of 90%. In the simultaneous inoculation, the results were compared with the S. aureus colony-forming units per milliliter (CFU/mL) count, as S. Typhimurium requires absence/presence rather than a count. It was found that in the UHT milk sample inoculated with S. Typhimurium and S. aureus, a higher count was observed than in raw milk, indicating that the microbiota present in raw milk could exert an inhibitory effect on the growth of S. aureus. In contrast, UHT milk inoculated additionally with L. lactis had higher growth than in all inoculations, suggesting some type of interaction among the three microorganisms.eng
