dc.description.abstractEl objetivo de este estudio fue evaluar el efecto de la estimulación ovárica con gonadotropinas sobre a) la cantidad de folículos ováricos de 3-9 mm al momento de la aspiración folicular, b) la cantidad y calidad de complejos cumulus oocitos obtenidos por sesión de aspiración folicular, c) la capacidad de desarrollo de los mismos medida por la tasa de clivaje y de blastocistos, así como determinar si existe efecto de la raza de la donadora en las tasas de recuperación y viabilidad de oocitos obtenidos mediante aspiración folicular. Veintitrés donadoras de razas Brahman, Jersey, Gyr, Simmental y Holstein, fueron sometidas a cuatro tratamientos: G1) control (sin tratamientos farmacológicos), G2) sincronización de la onda folicular sin estimulación con gonadotropinas G3) sincronización con estimulación con FSH y G4) sincronización con estimulación con eCG, en un modelo factorial cruzado. Un efecto positivo (p<0.05) fue encontrado en términos del número de folículos disponibles al momento de la OPU en los grupos de tratamientos G2, G3 y G4 comparados con los animales del grupo control (G1). No se encontraron diferencias estadísticamente significativas entre estos tres grupos (Sincronización, FSH o eCG). La sincronización de la onda folicular, afectó positivamente la cantidad de los complejos cumulus oocitos (COC´s), tasas de clivaje y blastocistos. Adicionalmente, se encontró un efecto positivo de la raza sobre la población de folículos presentes al momento de la OPU, tasas de recuperación, número de COC´s recuperados, tasas de maduración, de clivaje y de blastocistos. En conclusión, se encontró un efecto positivo de la sincronización de la onda folicular y de la raza sobre la calidad de los oocitos obtenidos y parámetros de producción de embriones in vitro, lo cual justificaría su utilización en programas de producción de embriones in vitro, más no se encontró en efecto benéfico del uso de gonadotropinas (FSH o eCG) antes de realizar la OPU. dc.description.abstractThe objective of this study was to evaluate the effect of ovarian stimulation with gonadotropins over the number of ovarian follicles (3-9 mm), at the time of Ovum Pick Up (OPU), the quantity and quality of oocyte cumulus complexes obtained per OPU session, their developmental capacity measured by the cleavage rate and blastocyst production; Additionally the effect of donor breed on the recovery and viability rates of oocytes obtained by follicular aspiration was also evaluated. Twenty-three donors of Brahman, Jersey, Gyr, Simmental and Holstein breeds were subjected to four treatments: G1) control (without pharmacological treatments), G2) synchronization of the follicular wave without gonadotropin stimulation, G3) synchronization and FSH stimulation and G4) synchronization and eCG stimulation, in a crossed factorial model. A positive effect was found in terms of the number of follicles available at the time of OPU in G2, G3 and G4 treatments compared to control group, with no significant differences between these three groups (synchronization, FSH or eCG). Regarding the number of COC's obtained, cleavage and blastocyst rates, a positive effect of synchronization was found, but not in the animals stimulated with FSH or eCG. Additionally, a positive effect of breed was found in terms of follicular population present at OPU, number of COC's recovered, viable COC's, COC's recovery, maturation and cleavage rate as well as blastocyst production. In conclusion, a positive effect of follicular wave synchronization, was found in cows with regard to follicular population, cleavage rate and blastocyst production, which would indicate its use in vitro embryo production programs; there was no beneficial effect on the use of gonadotropins (FSH or eCG).eng
