Determinación de la actividad antibacteriana de los extractos y hemolinfa de larvas de Lucilia eximia (Diptera: Calliphoridae)

Resumen: Lucilia eximia es una especie neotropical, de importancia médica y forense que ha sido utilizada en la terapia larval como método de desinfección, desbridamiento y cicatrización de heridas ulcerativas en pacientes en la ciudad de Medellín. En el presente trabajo, mediante técnicas de ultrafiltración y cromatografía de alta resolución (HPLC) se realizó una búsqueda preliminar de compuestos con actividad antibacteriana, presentes en exosecreciones y hemolinfa de larvas de Lucilia eximia. La evaluación de la actividad biológica de las fracciones de exosecreciones obtenidas por HPLC indica la presencia de compuestos activos sobre bacterias gram positivas y gram negativas, con pesos moleculares en el rango de 3-10 kDa y menores a 3 kDa. No se encontraron diferencias significativas en cuanto a la inhibición del crecimiento de las bacterias tratadas con excreciones y secreciones (E/S) expuestas y sin exposición a patógenos. Se encontraron ocho fracciones activas en un rango de peso entre 3-10 kDa y tres de tamaño molecular menor a 3 kDa, en la hemolinfa de larvas expuestas a patógenos. Las cepas Gram positivas, son más susceptibles a la exposición tanto de hemolinfa como de E/S provenientes de larvas de L. eximia de tercer estadio.
Abstract: Lucilia eximia is a neotropical species of medical and forensic importance, which has been used in maggot therapy as a method of disinfection, debridement and ulcerative wound healing in diabetic patients in Medellín. In this thesis, by ultrafiltration techniques and high performance liquid chromatography (HPLC ) a preliminary search of compounds with antibacterial activity present in larval hemolymph and exosecretions of L. eximia was made. The evaluation of the biological activity of exosecretions's fractions obtained by HPLC, indicates the presence of active compounds on Gram positive and Gram negative bacteria, with molecular weights between 3 and 10 kDa and less than 3 kDa. No significant difference in growth inhibition was observed between bacterial strains treated with exosecretions previously exposed or not exposed to pathogenic bacteria. Eight active fractions were found in a range between 3 and 10 kDa and three 3-fractions lower than 3 kDa molecular weight, in the hemolymph of larvae exposed to pathogens. The Gram-positive strains are more susceptible to exposure to both of hemolymph as exosecretions from third instar larvae of L. eximia.
